The intracellular forkhead winged transcription factor Foxp3 (forkhead box P3) appears to be specifically expressed by naturally occurring Treg cells, particularly in mice, although in humans there is evidence of upregulation of Foxp3 in all T cells on activation. Foxp3 is required for the development and function of naturally occurring regulatory t cells (treg) and expression is sufficient to convert non-regulatory CD4+CD25T cells into cells with regulatory activity. Conversion of peripheral CD4+CD25 naive T cells to Foxp3+CD4+CD25 foxp3+ Treg cells can be induced by TGF-ß. In a murine asthma model, these TGF-ß-induced Treg prevented house-dust mite-induced allergic pathogenesis or infection pathogenesis in lungs. A single independent report has suggested that IL-4 and IL-13 also induce Foxp3+CD25+ Treg from CD4+CD25precursors. (more…)

Antihistamines have been shown to be effective in seasonal allergic rhinitis and chronic idiopathic urticaria. They improve quality of life scores, acute inflammatory response markers in atopic dermatitis asthma and symptom scores. Newer histamine H1-receptor antagonists may also have an antiplatelet-activating factor effect and are equally effective in seasonal allergic rhinitis. The antileukotrienes were developed in the 1980s. The first compounds of this novel class of anti-asthma drugs were registered in the second half of the 1990s. The mechanism of action of the cysteinyl leukotriene receptor antagonists (LTRAs) is based on counteracting the effects of cysteinyl leukotrienes at their receptor site (CysLT1 receptor) within the airways. There have been concurrent increases in the prevalence of obesity and asthma in recent years in New Zealand and other countries. Two cross-sectional surveys performed in 1989 and 2000 were used to test this association in children of mean age 11.7 years. BMI was calculated as weight/height 2 (kg/m 2 ) and obesity and overweight defined according to an international standard. Standard questions were used to measure the prevalence of asthma symptoms. Exhaled Leukotrienes In Asthma & Non-Asthmatic Adult Patients With Seasonal Allergic Rhinitis

Leukotrienes are present in increased amounts in exhaled breath condensate (EBC) in patients with asthma. So far, no data have been reported about leukotriene levels in non-asthmatic patients with Seasonal Allergic Rhinitis. The aim of the study was to find out whether the leukotriene levels in exhaled breath condensate were increased in the non-asthmatic adult patients with Seasonal Allergic Rhinitis both during and after the pollen season in comparison with healthy controls, and to assess changes in leukotriene levels after the pollen season. Twentynine non-asthmatic adult patients with Seasonal Allergic Rhinitis and 50 healthy non-smoking controls underwent measurement of exhaled leukotrienes in the exhaled breath condensate during and after the pollen season. Spirometry, skin-prick tests and total IgE were also evaluated. (more…)

Otitis Media With Effusion: Allergic Inflammation In Middle Ear & Upper Airway

Otitis media with effusion is a chronic inflammatory disease of the middle ear space characterized by the accumulation of fluid. Previous investigations have suggested that the immunopathological mechanism underlying the development of middle ear effusion in allergy patients largely involves the effects of Th2 mediators. The investigators set out to determine whether the middle ear compartment may be a component of the united airways in allergic disease by comparing the inflammatory profiles of the middle ear with those of the upper airway. Middle ear effusions, torus tubaris (Eustachian tube mucosa at the nasopharyngeal orifice) and adenoidal tissue biopsies were obtained from 45 patients undergoing simultaneous tympanostomy tube placement for Otitis media with effusion and adenoidectomy for adenoid hypertrophy.
